ตั้งอยู่ในบริเวณโรงเรียนสตรีมารดาพิทักษ์ ถนนสันติสุข ตำบลจันทนิมิตร เป็นโบสถ์ที่มีประวัติการก่อสร้างยาวนานถึง 275 ปี ครั้งแรกสร้างขึ้นในปี พ.ศ. 2254  บนฝั่งตะวันตกของแม่น้ำจันทบุรี โดยคุณพ่อเฮิ้ต โตแลนติโน และบรรดาคาทอลิกชาวญวน  จนถึงปี พ.ศ. 2377 ได้มีการย้ายมาสร้างบนฝั่งตะวันออกของแม่น้ำจันทบุรีอันเป็นสถานที่ตั้งในปัจจุบัน โดยมิได้มีการบันทึกถึงสาเหตุของการย้ายแต่ประการใด และในปี พ.ศ. 2446 ได้ก่อสร้างโบสถ์หลังปัจจุบันขึ้นให้มีขนาดใหญ่กว่าหลังเก่าเพื่อรองรับกับจำนวนคริสตศาสนิกชนที่เพิ่มมากขึ้น โดยมีลักษณะเป็นสถาปัตยกรรมตะวันตกแบบโกธิค ภายในตกแต่งด้วยกระจกสีที่เรียกว่า     สเตนกลาส เป็นภาพนักบุญต่าง ๆ หลังจากนั้นได้มีการบูรณะซ่อมแซมมาโดยตลอด จนถึงปี พ.ศ. 2528 ได้มีการจัดงานฉลองโบสถ์คาทอลิกจันทบุรีครบรอบ 75 ปี นับได้ว่าโบสถ์คาทอลิกแห่งนี้เป็นโบสถ์ขนาดใหญ่ที่มีความเก่าแก่และกล่าวกันว่ามีความงดงามมากที่สุดในประเทศ
