โรงเรียนโพธาวัฒนาเสนีตั้งขึ้นเมื่อวันที่ 21 มิถุนายน 2451 (ร.ศ. 127) ตามประกาศในราชกิจจานุเบกษา เล่มที่ 25 หน้าที่ 380 ซึ่งคัดลอกโดยนายสังเวียน รัตนมุง อดีตผู้พิพากษาศาลจังหวัดสมุทรสาคร มีใจความดังนี้
"ด้วยได้รับใบบอกมณฑลราชบุรีที่ 3/1590 ลงวันที่ 19 พฤษภาคม ร.ศ.127(พ.ศ.2451) ว่าโรงเรียน
วัดโพธาราม เมืองราชบุรี เดิมได้อาศัยวัดเป็นที่เล่าเรียน แต่เป็นที่คับแคบมาก ด้วยมีนักเรียนทวีมากขึ้น ครั้งเมื่อ

ปี 2479 ได้ก่อสร้างอาคารเรียนขึ้นอีกเป็นครั้งที่ 3 เนื่องจากอาคารเรียนเดิมชำรุด และคับแคบเกินกว่าที่จะรองรับปริมาณนักเรียนที่เพิ่มขึ้นอย่างรวดเร็วได้ การก่อสร้างครั้งนี้มีพระครูโพธาภิวัฒน์ (เต็ก) เจ้าอาวาสวัดโพธาราม เป็นผู้ดำเนินการโดยก่อสร้างอาคารเรียน 2 ชั้น ใช้เงินบริจาค 9,442.43 บาท และกระทรวงศึกษาธิการจัดสรรงบประมาณสมทบเป็นเงิน 6,000 บาท รวมราคาค่าก่อสร้างเป็นจำนวนเงิน 15,442.43 บาท
ปี 2483 โรงเรียนยังคงจัดสอนระดับประถมศึกษาอยู่ด้วย ภายหลังได้ทยอยตัดออกไปจนเหลือแต่เพียงระดับมัธยม ซึ่งเปิดสอนชั้นมัธยมปีที่ 1 ถึงชั้นมัธยมปีที่ 4 และได้เปิดสอนถึงชั้นมัธยมศึกษาปีที่ 6 เป็นปีแรก นับเป็นโรงเรียนมัธยมระดับอำเภอแห่งแรกในประเทศไทย ที่เปิดสอนถึงชั้นมัธยมศึกษาปีที่ 6
