มหาอำมาตย์ตรีพระยาเพชรพิไสยศรีสวัสดิ์ จึงขอพระราชทานเงินบริจาคดังกล่าว มาสร้างเป็นโรงเรียนสตรีประจำจังหวัดโดยพระบาทสมเด็จพระมงกุฎเกล้าเจ้าอยู่หัว ทรงพระราชทานนามโรงเรียนว่า เบญจมเทพอุทิศ เพื่ออุทิศทูลเกล้าถวายแด่พระบาทสมเด็จพระจุลจอมเกล้าเจ้าอยู่หัว โรงเรียนจึงมีชื่อเดิมว่า โรงเรียนสตรีเพชรบุรีเบ็ญจมเทพอุทิศ เปิดครั้งแรก เมื่อวันที่ 1 สิงหาคม พ.ศ. 2460
ใน พ.ศ. 2473 สมเด็จพระเจ้าพี่นางเธอเจ้าฟ้าวไลยอลงกรณ์ กรมหลวงเพชรบุรีราชสิรินธร ได้พระราชทานเงินส่วนพระองค์จำนวน 6,000 บาท เพื่อรื้ออาคารเดิม และก่อสร้างอาคารใหม่ที่เดิม นอกจากนี้ยังพระราชทานเงินส่วนพระองค์จำนวน 2,000 บาท เพื่อจัดซื้ออุปกรณ์เพิ่มเติม
ใน พ.ศ. 2498 โรงเรียนย้ายมา ณ ที่ตั้งปัจจุบัน อันเป็นที่ดินทรัพย์สินส่วนพระมหากษัตริย์ เนื้อที่ 36 ไร่ 1 งาน 76 ตารางวา และใช้ชื่อใหม่ว่า โรงเรียนเบญจมเทพอุทิศจังหวัดเพชรบุรี
