วัดธาตุ พระอารามหลวง ตั้งอยู่เลขที่ ๒๓๗ ถนนกลางเมือง (บ้านเมืองเก่า) ตำบลในเมือง อำเภอเมืองขอนแก่น จังหวัดขอนแก่น สังกัดคณะสงฆ์มหานิกาย สร้างขึ้นเมื่อ พ.ศ. ๒๓๓๒ โดยพระนครศรีบริรักษ์บรมราชภักดี (เพี้ยเมืองแพน) เจ้าเมืองขอนแก่น เป็นผู้ก่อสร้างขึ้นพร้อมกับการตั้งเมืองขอนแก่น และได้สร้างพระธาตุขึ้นเป็นปูชนียสถานที่เคารพสักการะของชาวเมืองขอนแก่น จึงเรียกชื่อวัดนี้ว่า "วัดธาตุ" ได้รับการสถาปนาขึ้นเป็นพระอารามหลวง ชั้นตรี ชนิดสามัญ เมื่อปี พ.ศ. ๒๕๒๑ มีเนื้อที่ ๑ ๒ไร่มีอาณาเขตดังนี้

เมื่อ พ.ศ.๒๕๑๓ คณะกรรมการวัดเห็นว่าพระธาตุมีรวมกันหลายองค์ ทุกองค์ล้วนมีลักษณะทรุดโทรม จนไม่สามารถทราบได้ว่าองค์ไหนเป็นพระธาตุองค์เดิม จึงได้ตัดสินใจให้นายช่างออกแบบสร้างพระธาตุองค์ใหม่ขึ้นครอบองค์เดิมไว้ ทั้งหมด โดยสร้างให้ใหญ่และสูงกว่าเดิม เพื่อรักษาศรัทธาปสาทะและความศักดิ์สิทธิ์ไว้ และให้ชื่อว่าเจดีย์พระธาตุนครเดิม ตามที่ปรากฏอยู่ทุกวันนี้ มีส่วนสูงวัดจากยอดสุดลงมา ๔๕ เมตร
พ.ศ. ๒๔๓๐-๒๔๗๓ พระครูพุทธา พุทธฺสโร เจ้าอาวาส การปกครองในยุคนี้เป็นระเบียบเรียบร้อย มีพระภิกษุ สามเณรมาก มีการศึกษาพระธรรมวินัย เรียกในสมัยนั้นว่า เรียนสนธิ เรียนนาม เรียนมูลกัจจายน์ และมีการส่งเสริมการเทศน์เสียงลำมหาชาติทำนองพื้นเมือง พระครูพุทธา เจ้าอาวาส เป็นผู้ทรงความรู้ความสามารถในการเทศน์ทำนองเสียงลำมหาชาติพื้นเมืองชั้นหนึ่ง ถึงมรณภาพในปี ๒๔๗๓
พ.ศ. ๒๔๓๐ - ๒๔๘๓ พระครูบับ ปญฺญาวโร ศิษย์ของพระครูพุทธา ได้เป็นเจ้าอาวาส ต่อมาท่านได้ปกครองวัดเจริญรอยตามพระครูบาอาจารย์ทุกอย่าง พระสงฆ์ สามเณร และประชาชนเคารพนับถือมาก เช่น เมื่อ พ.ศ. ๒๔๗๕ ได้มีศรัทธาสร้างกุฎีถวายพระครูบับสำหรับเป็นที่อยู่ของเจ้าอาวาสในสมัยนั้น (ขณะนี้รื้อไปแล้ว) พระครูบับ ปญฺญาวโร เป็นเจ้าอาวาสอยู่ประมาณ ๑๐ ปี ได้ลาสิกขาไปเมื่อปลายปี พ.ศ. ๒๔๘๓
พ.ศ. ๒๔๘๓ - ๒๔๙๐ พระครูวิเวกธรรมปฏิบัติ (พั้ว พุทฺธโชโต) เจ้าคณะแขวงพระลับ เจ้าอาวาสวัดท่าราชไชยศรี ได้รับความเห็นชอบจากทางคณะสงฆ์และทายก-ทายิกา ให้ย้ายมาดำรงตำแหน่งเจ้าอาวาสวัดธาตุ ท่านได้ปรับปรุงการปกครอง การศึกษาการเผยแผ่ และการสาธารณูปการโดยกว้างขวาง ซึ่งนับเป็นรากฐานของวัดธาตุสืบมาจนบัดนี้ ทั้งนี้เพราะท่านเป็นทั้งนักปกครอง นักศึกษา นักเทศน์ และนักก่อสร้าง
พ.ศ. ๒๔๙๐ – ๒๕๕๒ พระธรรมวิสุทธาจารย์ (เหล่ว สุมโน ป.ธ. ๕, พธ.ด.) เป็นเจ้าอาวาส ได้มีการปรับปรุง พัฒนาทั้งด้านการศึกษา การปกครอง การเผยแผ่ และสาธารณูปการเต็มรูปแบบ ซึ่งในอดีตนั้นวัดธาตุมีการจัดการศึกษาเล่าเรียนตามแบบพื้นเมืองโบราณ ถ้าพระภิกษุสามเณรรูปใดสนใจใฝ่ศึกษาจะต้องเดินทางไปศึกษาเล่าเรียนที่กรุงเท ฯ จนกระทั่งสมัยพระครูวิเวกธรรมปฏิบัติ (พั้ว เป็นเจ้าอาวาส ได้วางรากฐานการศึกษาพระปริยัติธรรม แผนกนักธรรม-บาลีขึ้น โดยได้สร้างโรงเรียนพระปริยัติธรรมและจัดหาครูสอนพระปริยัติธรรมจากวัดมหาธาตุ กรุงเทพฯ ๒ รูป คือ พระมหาเหล่ว สุมโน ป.ธ.๕ (พระธรรมวิสุทธาจารย์) และพระมหาพราม จิตฺตเสโน ป.ธ.๖ มาเป็นครูสอนประจำสำนักรุ่นแรก ต่อมาเมื่อถึงสมัยพระธรรมวิสุทธาจารย์ (เหล่ว สุมโน) เป็นเจ้าอาวาส ได้ส่งเสริมและปรับปรุงพัฒนาสำนักเรียนให้เจริญก้าวหน้ามากยิ่งขึ้น โดยแต่ละปีมีพระภิกษุสามเณรเข้าศึกษาและสอบเปรียญธรรมได้เป็นจำนวนมาก มีครูสอนพระปริยัติธรรมที่มีความรู้ความสามารถทำการสอนประจำในอดีต ได้แก่
