บทคัดย่อ
​ในการศึกษาทดลองเรื่อง การปนเปื้อนของเชื้อ Coliform Bacteria ในน้ำดื่มของโรงเรียนจุฬาภรณราชวิทยาลัย ลพบุรี มีวัตถุประสงค์เพื่อตรวจสอบคุณภาพทางจุลชีววิทยาของน้ำดื่มในโรงเรียนจุฬาภรณราชวิทยาลัย ลพบุรี และเพื่อตรวจปริมาณโคลิฟอร์มและ E. coli ในน้ำดื่ม รวมทั้งน้ำแข็ง ตามจุดบริการต่างๆที่นักเรียนในโรงเรียนใช้บริโภค ดำเนินการโดยแบ่งเป็น 3 ขั้นตอน ได้แก่
ขั้นตอนที่ 1 การทดสอบขั้นแรก (Presumptive test) ในการทดสอบใช้ระบบเลี้ยงเชื้อแบบ 3 หลอดกับอนุกรม 3การเจือจาง คือ จำนวนมิลลิลิตรของตัวอย่างที่ต่างกันเป็นชุดๆ ดังนี้ 10 - 1 - 0.1 มิลลิกรัมต่อลิตร
ขั้นตอนที่ 2 การทดสอบขั้นยืนยัน (Confirm test) เนื่องจากการทดสอบขั้นแรกไม่สามารถชี้ชัดว่าแบคทีเรียที่อยู่ในตัวอย่างน้ำเป็นโคลิฟอร์มแบคทีเรีย หรือฟีคัลโคลิฟอร์มแบคทีเรีย ดังนั้นจึงต้องทดสอบยืนยันโดยการถ่ายเชื้อลงในอาหารเลี้ยงเชื้อ Brilliant – Green Lactose bile broth และ EC medium
ขั้นตอนที่ 3 การทดสอบขั้นสมบูรณ์ (Completed test )นำเชื้อที่เกิดฟองอากาศในขั้นยืนยันมาเขี่ยเชื้อลงบนจานเพาะเชื้ออาหารแข็ง Eosin Methylene Blue agar (BMB)
ผลการทดลองพบว่า ในน้ำตัวอย่างทั้ง 3 ตัวอย่างพบว่า
1.ค่าโคลิฟอร์มแบคทีเรีย

​น้ำดื่มในโรงอาหาร 93 MPN/100 mL
​น้ำกรองบนหอพัก 9 MPN/100 mL
2.ค่าฟีคัลโคลิฟอร์มแบคทีเรีย
​น้ำแข็งในโรงอาหาร >2400 MPN/100 mL
​น้ำดื่มในโรงอาหาร 93 MPN/100 mL
​น้ำกรองบนหอพัก 9 MPN/100 mL
