การเดินทางเข้าสู่อุทยานแห่งชาติเขาสกมีสองเส้นทาง คือ จากอำเภอเมืองสุราษฎร์ธานี หรืออำเภอตะกั่วป่า จังหวัดพังงา ตามเส้นทางหลวงแผ่นดินหมายเลข 401 (สุราษฎร์ธานี-ตะกั่วป่า) แยกเข้าอุทยานแห่งชาติเขาสก ตรงหลักกิโลเมตรที่ 109 ไปอีกประมาณ 1.5 กิโลเมตร ถึงที่ทำการอุทยานแห่งชาติ
